Substrates of luciferase

Luciferase與其substrate催化反應後能夠產生出可偵測的冷光訊號，是常用與Reporter assays的實驗方法之一。最代表的研究應用為gene expression實驗，我們能在想偵測的target gene後面加上reporter gene (luciferase DNA sequence)。最後使用Luciferase與substrates反應所產生冷光的訊號，來偵測target gene表達的位置及表達量。我們的substrate除了有最常見的D-Luciferin (particularly firefly luciferin)。同時也有Coelenterazine，可用於偵測細胞內鈣離子濃度、超氧離子及其他需要生物冷光的應用。

1.  Flah Type -Luciferase Assay System

無需反應等待時間，適合即加即測的實驗或是冷光訊號較低的樣品需增加靈敏度的實驗

2.  Bright Type -Luciferase Assay System

在短時間內即可達到放光極大值並維持放光，適合high troughput applications 及需要增加靈敏度的實驗

3.  Steady Type -Luciferase Assay System

Steady type 訊號強度較低但可穩定持續長時間放光，適合需要長時間紀錄冷光訊號的實驗及 high throughput application使用
